-April 10,2021 selected unknown ID#57
-April 18, 2021 requested the gram stain result for #57
-April 19, 2021 received gram stain image:
It is rod shaped and purple in color, making it a grampositive.
-April 23, 2021 my unknown is a gram-positive rod, and attached is the gram-positive rod dichotomous
key.
-May 5 , 2021 requested the results for a catalase test of Unknown#57
-May 5, 2021 received catalase test result:

Results show that it is positive as it produces bubbles. Since the results are
positive I move on to the next step which is to request an endospore stain.
-May 6, 2021 requested and received endospore stain image:
Image shows that it produces endospores, as they are green
while the vegetative cells remain red. From this I am able to
conclude that my group ID is Bacillus.
Determining the species of my group ID
-Revisited the catalase test from the time I was determining my group ID: It is a positive result so I
eliminated 2 species leaving me with 93 species.
-May 16, 2021 requested growth at pH 6 and 7
-May 17, 2021I received the image below:
Cloudy color, based on my observation there is growth present making it positive. This automatically
disqualifies 26 species, leaving me with 67 species.
-May 18,2021 requested and received growth at 20 degrees Celsius and 40 degrees Celsius :

Cloudy color indicates presence of growth making it a positive result. This eliminates 11
species, leaving me with 56 species.
-May 19, 2021 requested growth in NaCl at 7% and 10%
-May 20,2021 received image:
At 10% Clear tube indicates a negative test, from this I can eliminate 12 species,
leaving me with 44 species.
At 7% Indicates growth as tube is cloudy, so making it a positive result. For this we can eliminate 11
species, leaving me with 33 species.
-June 3, 2021 requested motility test and growth at 50 degrees Celsius
-June 4, 2021 received images:
Growth at 50 degrees shows a clear tube which indicates that test is negative. From this, I can
eliminate 13 species, leaving me with 20 species.

Motility test: Indicates a thin pink line which makes it a positive test, for this I can eliminate 3 species,
leaving me with 17 species.
– June 7, 2021 requested hydrolysis of starch and hydrolysis of casein
– June 7, 2021 received images:
For hydrolysis of starch, there is glow around growth which indicates a positive
result, for this I can eliminate 1 specie, leaving me with 16 species
For hydrolysis of casein, there is a clear zone around colonies making it a positive
result. For this, I will eliminate 6 species, leaving me with 10 species.
– June 9, 2021 requested acid from glucose and acid from mannitol
– June 10,2021 received images:
For mannitol, red color indicates a negative color with no acid present. So I will eliminate 4
species, leaving me with 6 species.

For Glucose, color is yellow which indicates a positive result. So I will eliminate 2 species,
leaving me with 4 species.
– June 11, 2021 requested nitrate reduction, utilization of citrate, and voges-proskauer
– June 11, 2021 received images:
For Voges-proskauer, brownish red color at the top of the tube indicating a positive result. For
this I eliminate 2 species leaving with 2 species.
For nitrate-step 1, red color indicates a positive result. So I eliminate 1 specie leaving me with 1
species . For this I can conclude that my species is B.subtilis, my Unknown is a bacillus subtilis.
-Utilization of citrate test was inconclusive
